p16和p53在食管鳞状细胞癌中的表达及意义 被引量:15

摘要 目的 探讨 p16、p53在食管鳞状细胞癌中的表达及临床意义。方法 应用 PCR和 PCR-SSCP法。结果  p16缺失率在食管鳞状细胞癌 a期与 b期、 期中分别为 45.0 %与 92 .3 %、10 0 .0 % ,在高分化组与中、低分化组中分别为 9.1%、52 .2 %和 10 0 .0 %。 p53突变率在鳞状细胞癌组织和正常黏膜组织中分别为 46.7%和 0 ,两者差异有显著性 ;在高、中分化组与低分化组中分别为 2 0 .0 %、2 1.4%与 90 .9% ,前两者与后者相比差异有显著性。结论 食管鳞状细胞癌存在着显著的 p16缺失和 p53突变。 p16的缺失与肿瘤的病程进展有关 ;p53的缺失突变与肿瘤的病程进展无关 。 ObjectiveTo investigate the relationship between loss of pl6, mutation of p53 and squamou s cell carcinoma of the esophagus. MethodsForty cases with esoph ageal cancer and 20 cases of normal esophageal mucous membranes waer included in the study. p16 gene in the tissues was detected using PCR technology and p53 g ene was detected using PCR-SSCP silver staining technology. ResultsThe rates of p16 gene loss in the squamous cell carcinoma and normal mucos a tissues of esophagus were 47 5% and 0 (P<0 01). In the patients with or without lymph node metastasis the p16 loss rates were 72 7% and 37 9% (P< 0 05); in stage Ⅱa, Ⅱb, Ⅲ the rates were 45 0%,92 3% and 100 0%, respect ively (P< 0 01 ); in carcinoma with high, middle and low differentiation the rates were 9 1% and 52 2% and 100 0%, respectively (P< 0.01 ). But the p16 loss rate was not associated with the age and sex of patients, and site s of tumor. The mutation rates of p53 gene in the squamous cell carcinoma and no rmal mucosa tissues of esophagus were 46 7% and 0 (P<0 05); in carcinoma with high, middle and low differentiation the rates were 20 0%, 21 4% and 90 9%, respectively (P<0 05). The p53 mutation rates in the exon 5,6,7,8 wer e 13.3%,6.7%,16.7% and 10.0%, respectively (P>0 05), but those were associ ated with the age and sex of patients, and TNM stage and site of tumor. ConclusionsThe loss of p16 and the mutation of p53 is a common event i n the squamous cell carcinoma of esophagus and may play an important role in the development of carcinoma.
