涂敷担体和柱温对纤维素三苯基氨基甲酸酯涂敷型固定相拆分奥美拉唑的影响 被引量:2

摘要 分别用氨丙基硅胶和不改性硅胶作涂敷担体,制备了纤维素三苯基氨基甲酸酯(TPCC)涂敷型手性固定相,用乙醇/正己烷作流动相,对奥美拉唑进行了手性拆分,并考察了温度对拆分效果的影响,发现用不改性硅胶作担体,亦可取得较好的拆分效果.容量因子和分离因子随柱温升高而降低.分别用ChiralpakAD柱和自制的TPCC手性柱分析有一定对映体过量值(ee)的奥美拉唑溶液,确定了奥美拉唑两对映体在TPCC涂敷型固定相上的洗脱顺序. Chiral stationary phases(CSPs) were prepared by coating cellulose triphenylcarbamate (TPCC) on macropore aminopropyl silica gel(APS) and unmodified silica gel. Enantiomers of omeprazole were resoluted on the two different CSPs using ethanol/hexane as mobile phase. It was found that the unmodified silica gel can be used as a coating support, but the resolution on CSPs based on it is a little poorer than that on CSPs based on APS. The effect of column temperatures on the resolution was investigated and the capacity factor and separation factor of enantiomers of omeprazole decrease as the column temperature increases. The elution order of enantiomers of omeprazole on TPCC coated CSPs was determinded , and the (S)-enantiomer was eluted firstly.
