摘要
M胆硷受体上行调节具有以下特点:(1)用100nmol/L的人工合成的糖皮质激素地塞米松(dexamethasone,DEX)孵育细胞48h,细胞中M受体量从726.0±25.9增加至1092.0±39.0fmol/10~6细胞,DEX不影响M受体与配基结合的亲和力.(2)DEX的调节作用在一定范围内依赖于时间和浓度,10^(-8)mol/L的DEX达最大效应.(3)DEX可使细胞对标记前体向DNA、RNA和蛋白质的参入量分别增加80.8±5.1%,29.8±3.3%和20.0±3.2%,表明DEX对M胆硷受体的调节作用与促进DNA复制、RNA砖录和受体蛋白质的合成过程有关.
Treatment of CCL137 cells with 100 nmol/L dexamethasone(DEX) for 48 hours caused increase in the number of muscarinic acetylcholine receptor (mAChR) from 726.0±25.9 to 1092.0±39.0 fmol/10~6 cells,without affecting its affinity to ligand.The effect of DEX on mAChR was dose-and time-dependent.The maximum response was induced by 10^(-8)mol/L of DEX,The incorporation of tritiated-thymidine,uridine and leucine into DNA,RNA and protein of the cells was increased by DEX to 80.8±5.1,29.8±3.3 and 20.0±3.2% respectively as compared to the control's.This means that the effect of DEX on mAChR may be due to the enhancement of DNA replication,RNA transcription and receptor protein synthesis.
