糜蛋白酶应用于生物检材中地西泮含量的测定 被引量:1

摘要 目的 :测定生物检材中地西泮的含量 ,提高地西泮的提取率。方法 :兔肝经糜蛋白酶水解、固相萃取法提取后 ,用气相色谱 质谱联用仪分析地西泮的含量。结果 :酶解后的肝脏中地西泮的检出量明显增加 ,每克肝组织添加 1 0 0 0u糜蛋白酶时效果最佳。结论 :糜蛋白酶的应用提高了生物检材中地西泮的提取率。 OBJECTIVE To determine the contents of diazepam in biological samples and to improve its rate of extraction. METHODS The samples were hydrolyzed by chymotrypsin of different quantity, and diazepam was extracted by solid-phase columns and its contents were analyzed by gas chromatography-mass spectrometry(GC-MS).RESULTS It is shown that the contents of diazepam increased when the amount of chymotrypsin was increased in the sample. CONCLUSIONS This method improved the extraction rate of diazepam in biological samples.
