摘要
本文报道了一个用离子配对高效液相色谱测定哺乳动物细胞内尼克酰胺腺嘌呤二核苷酸(NAD)含量的新方法.这个方法与1981年Jones法相比,具有以下明显的优点:(1)NAD峰分离清楚,无拖尾现象;(2)出峰早,节省操作时间,适合于多样品操作;(3)单泵洗脱,不需梯度仪,设备比较简单;(4)洗脱液对泵和色谱柱无不利影响.本法快速简便,灵敏度较高,结果重现好.
A new method for the determination of nicotinamde adenine dinucleotide (NAD) in mammalian cells by ion-pair HPLC is reported.In comparison with the HPLC method of Jones (1981),this method has several advantages,such as good separation,early appearance of NAD peak,no harmful effect of the eluting solution to the pump and column,and simpler installments.This method is simple,rapid,and accurate.As little as 25ng of NAD can be detected.
