内皮素-1受体在兔不全梗阻性膀胱中的研究

摘要 目的 :研究内皮素 1受体 (ET 1R)在兔不全梗阻性膀胱中的变化和意义。 方法 :采用受体的放射配基结合分析技术 (RBA) ,对 10只成年雄性新西兰白兔不全梗阻性膀胱 (兔不全梗阻性膀胱模型 ,梗阻 6周为实验组 )及 10只同龄雄性新西兰白兔无梗阻性膀胱术后 6周 (对照组 )的膀胱体及膀胱颈部平滑肌组织ET 1R进行研究。结果 :在膀胱体平滑肌组织中ET 1R结合量 ,实验组较对照组明显增多 (P <0 .0 2 ) ,而膀胱颈平滑肌组织中ET 1R结合量无明显变化 (P >0 .2 )。实验组较对照组的膀胱重量明显增加 (P <0 .0 0 1)。 结论 :兔不全梗阻性膀胱的膀胱体平滑肌组织中ET 1R结合量明显增多。ET 1具有收缩平滑肌和促平滑肌、纤维结缔组织增生 (有丝分裂 )的作用 ,故ET 1R的上调是导致兔不全梗阻性膀胱壁结构改变及功能紊乱的重要因素之一。 Objectives : To study endothelin 1 receptor in a rabbit model partial bladder outlet obstruction(BOO). Methods :We used the rabbit model of partial bladder outlet obstruction to study endothelin 1 receptor(ET 1R) with radioligand binding assay of receptors (RBA).Partial bladder outlet obstruction was induced in adult male New Zealand white rabbits(ten rabbits as experiment group,time of obstruction for six weeks).Sham operated age matched rabbits acted as control(ten rabbits as control group,time of postoperation for six weeks). Results: The endothelin 1 binding sites were significantly increased in the BOO detrusor smooth muscle( P <0.02).However, there was no changes in the BOO bladder neck . BOO bladder weights were significantly increased( P <0.001). Conclusions: The endothelin 1 binding sites were significantly increased in the BOO detrusor smooth muscle . Endothelin 1 has mitogenic and potent contractile properties for smooth muscle cells,has mitogenic property for connective tissue.This suggests the up regulation of ET 1R is associated with altered bladder structure and disfunction of rabbit partial bladder outlet obstruction .
