摘要
目的 探讨灵芝对糖尿病大鼠早期肾病的干预作用。 方法 4 3只正常雄性SD大鼠 ,分成正常对照组 (C组 )和糖尿病组 (系链脲佐菌素诱发糖尿病大鼠动物模型 )。糖尿病组随机分成糖尿病对照组 (D组 ) ,苯那普利治疗组 (A组 ) ,灵芝治疗组 (L组 )。A组予苯那普利 10mg·kg-1·d-1灌胃 ,L组予灵芝 6 0 0mg·kg-1·d-1灌胃 ,D组予相同体积的饮用水灌胃。 结果 治疗 12周后 ,L组和A组的肾重 /体重、尿白蛋白排泄率、肾基质 /肾小球面积比较D组明显下降 ;D组肾皮质的转化生长因子 β1(TGF β1)mRNA的表达增强 ,A组和L组较D组减弱 ,TGF β1免疫组化染色也同步改变。 结论 灵芝能明显降低糖尿病鼠的尿微量白蛋白的排泄率及形态学异常 ,并在某种程度上抑制了TGF β1的表达和合成 。
Objective The aim of the present study was to investigate the effect of Lingzhi on the early nephropathy of diabetic rats. Methods Forty three SD male rats were randomly divided into normal (group C) and diabetes rats which were induced with peritoneal injection of streptozocin. The latter were randomly divided into diabetes group (group D), Benazepril treatment group (group A) and Lingzhi treatment group (group L). Group A was treated with Benazepril (10 mg·kg -1 ·d -1 , group L with Lingzhi(600 mg·kg -1 ·d -1 ),and group D (without any treatment was given water through tube feeding. Result After 12 weeks treatment, the renal weight/body weight and the urinary albumin excretion rate(UAER) in group L and A were significantly decreased compared with group D; the ratio of ECM area to glomerular area was reduced in group A and L. The expression of transforming growth factor β 1 (TGF β 1) mRNA was higher in group D, but it was significantly reduced in group A and L and and the type Ⅳ collagen and TGF β 1 immunohistochemical staining showed the corresponding changes in group A and L compared with group D. Conclusion Lingzhi has a significant effect of decreasing UAER, and also a protected role by the down regulated expression and synthesis of TGF β 1 in the early nephropathy of diabetic rats.
