摘要
目的 探讨肺内肿瘤坏死因子 α(TNF α)在大鼠急性坏死性胰腺炎 (ANP)所致肺损伤中的作用。方法 将大鼠分为正常对照、ANP和TNF α抗体预处理 3组 ,后两组又分为 1、3、6、12h 4组 ,每组 6只。胰胆管注入 3 %牛磺酸钠诱发ANP。肺灌洗获得巨噬细胞 ,培养后检测TNF α水平、肺泡灌洗液 (BALF)中蛋白含量 ,取肺组织测髓过氧化物酶 (MPO)水平。逆转录 聚合酶链反应 (RT PCR)检测巨噬细胞TNF αmRNA表达。结果 ANP和TNF α抗体预处理组各指标均较正常对照升高 (P <0 .0 5 )。TNF α水平与MPO及BALF蛋白含量呈正相关 (P <0 .0 1) ,TNF αmRNA的表达及TNF α水平与肺损害程度呈正相关 (P <0 .0 1)。结论 肺泡巨噬细胞TNF α的过表达与ANP大鼠所致肺损伤有密切关系。
Objective To investigate the role of tumor necrosis factor-α(TNF-α) secreted by alveolar macrophages (AM) in lung injury associated with acute necrotizing pancreatitis (ANP).Methods Rats were randomly divided into three groups:normal control,ANP and TNF-α antibody pretreatment.Each group was divided into 1,3,6 and 12?h subgroups (n=6 in each group).Rat ANP model was induced by intraductal administration of 3% sodium taurocholate.AM were obtained by bronchoalveolar lavage,then cultured for 24 h.The protein content of bronchoalveolar lavage fluids (BALF),the myeloperoxidase (MPO) of lung tissue and generation of TNF-α were evaluated.The expression of TNF-α?mRNA was detected by reverse transcription-polymerase chain reaction.The histology of lung was also checked.Results The lung injury was gradually aggravated.The level of MPO,protein content and generation of TNF-α were also elevated in a time-dependent manner.The expression of TNF-α?mRNA was gradually upregulated after the onset of ANP and reached their peaks at 6 h.There was statistical significant difference in all the indicators between ANP and TNF-α antibody pretreatment groups and normal control group (P<0.05).The generation of TNF-α had a close relation to the level of MPO of lung tissue and protein content of BALF (P<0.01).The expression of TNF-α?mRNA and the generation of TNF-α had a close relation to the lung injury (P<0.01).Conclusion The overexpression of TNF-α secreted by AM may play an important role in lung injury associated with ANP.
