摘要
目的 观察硒对人脐静脉内皮细胞 ECV- 30 4单核细胞趋化蛋白 - 1(MCP- 1)表达的影响。方法 分别用高葡萄糖、糖基化终末产物 ,高胰岛素和过氧化氢孵育人脐静脉内皮细胞 ECV- 30 4 ;在先加入 10 0 nmol/L硒后 ,再给予以上 4种因素 ,分别检测人脐静脉内皮细胞 ECV- 30 4 MCP- 1m RNA的表达并比较。结果 4种因素均可作为独立因素 ,导致人脐静脉内皮细胞 ECV- 30 4 MCP- 1m RNA表达量增加 ;硒能抑制 4种因素所致的MCP- 1m RNA表达。结论 硒抑制 MCP- 1m RNA在人脐静脉内皮细胞 ECV- 30 4中的表达。
Objective To observe the effect of selenium on expression of monocyte chemoattractant protein-1 (MCP-1) mRNA in human umbilical vein endothelial cell line ECV-304.Methods Cell line ECV-304 was incubated with high glucose, advanced glycosylation end products (AGE), high insulin and H 2O 2 respectively;simultaneously, the cell line ECV-304 pre-treated with selenium was incubated with above factors, the expression of MCP-1 mRNA was detected and compared.Results Four factors increased the expression of MCP-1 mRNA independently;selenium inhibited the expression of MCP-1 mRNA induced by 4 factors distinctly.Conclusions Selenium repress the expression of MCP-1 mRNA in the cell line ECV-304.
