摘要 目的 观察Cx4 3基因异常引起心功能障碍的超声心动图表现。方法 对胎龄为 14 5d、15 5d或16 5d的Cx4 3基因剔除、CMV4 3和FZ转基因小鼠胚胎 ,进行多普勒超声心动图检测、心脏形态学检查和基因型测定。结果 Cx4 3基因异常小鼠胚胎主要表现为心室收缩期射血峰值增高 (IPSV)或 和舒张期房室瓣口血流频谱呈单峰 (MIP)。IPSV增高与肺动脉口狭窄相一致 ;而舒张期MIP则与右心室肥厚或右心室扩大相符合。结论 超声心动图在小鼠心脏病模型的研究方面具有较高的价值 ;Cx4 3异常可导致肺动脉口狭窄和右室舒张功能障碍。 Objective To investigate the echocardiographic features of the cardiac abnormalities in the mouse embryos with Cx43 gene defects. Methods Three mouse lines were used in the study, which included Cx43 knockout mice (Cx43KO), transgenic mice overexpressing Cx43 (CMV43) and transgenic mice expressing Cx43/β-galactosidase fusion protein (FZ). In-utero Doppler echocardiography and the pathological analyses were carried out to examine the embryonic cardiac abnormalities at embryonic day (ED) 14.5, ED15.5 or ED16.5. Results The studies showed that cardiac defects in Cx43KO, CMV43 and FZ mice were mainly associated with the obstruction of right ventricular outflow tract and abnormal myocardial development in presentation of the increased peak systolic ejection velocity (IPSV) and the monophasic diastolic inflow pattern respectively. Echocardiographic findings were consistent with the morphological changes including conotruncal pulmonary stenosis and right ventricular hypertrophy or enlargement. Conclusion Doppler echocardiography is of importance in the study of heart defects in mice with the gain or loss of Cx43 function,which lead to the obstruction of pulmonary outflow tract and disorder of diastolic function of right ventricle.
