摘要
目的 :探讨锌对人类精子受外源性一氧化氮 (NO)供体硝普钠 (SNP)损伤的保护性作用 .方法 :利用相差显微镜及单细胞凝胶电泳分析锌对SNP影响精子运动能力、精子细胞头尾连接及精子细胞DNA链断裂的保护性 .结果 :SNP对精子运动能力有明显的抑制作用 ,精子彗星细胞百分率显著增加 ,精子头尾连接稳定百分率降低 ,而锌离子能提高精子的运动功能 ,防止精子细胞DNA链的断裂和精子头尾连接的解离 .结论 :锌对NO损伤精子有一定的保护作用 ,其机制可能与精子核染色质部位的巯基有关 。
AIM: To discuss the preventive function of zinc on human sperms from being injured by sodium nitroprusside (SNP), an external NO donor. METHODS: Phase contrast microscope and single cell gel electrophoresis (SCGE) were used to analyze the function of zinc in relieving sperms from influences given by SNP, such as the moving capability of sperms, the head tail connection of sperm cells, the break apart of sperm cells DNA chain. RESULTS: SNP inhibited the moving capability of sperm cells obviously, increased the percentage of comet cells obviously, and decreased the percentage of the head tail connection stability of sperm cells. But all these influences were relieved by zinc.CONCLUSION: Zinc can protect sperm cells from being injured by NO. The mechanism may be related to the mercaptol group of sperm chromatin. In a word, zinc has important value in the therapy of male sterile.
