摘要 目的 从细胞水平上探讨8-Br-cAMP对视网膜母细胞瘤株增殖的影响。方法 采用8-Br-cAMP对视网膜母细胞瘤株HXO-Rb_(44)细胞进行体外实验,通过蛋白斑点印迹方法检测HXO-Rb_(44)细胞PCNA-IR的OD值,观察其增殖情况,流式细胞术检测细胞周期变化。结果 HXO-Rb_(44)细胞增殖受抑制,PCNA OD值与药物作用时间呈显著负相关(P<0.01),且细胞周期发生改变,被阻滞于G_0/G_1期,S期比率下降,亦与药物作用时间有关。结论 8-Br-cAMP具有抑制Rb_(44)细胞增殖作用,使其阻滞于G_1期,为临床治疗视网膜母细胞瘤提供了新的途径。 Objective To clarify the effect of 8-Br-cAMP on proliferation of HXO-Rb44 cells. Methods HXO-Rb44 cells in vitro were treated with 2 x 10-5 mol/L 8-Br-cAMP for 24 h ,48 h, and 72 h. The proliferative cell nuclear antigen ( PCNA ) was observed by protein dot blot, and the cell cycle was detected by flow-cytometry. Results HXO-Rb44 cell growth was inhibited by 8-Br-cAMP. The difference between experiment group and control group was significant( P < 0. 01). There was a negative correlation between PCNA OD value and 8-Br-cAMP active time. The effects of 8-Br-cAMP on ratio of G0/G1 and S phases changed with the time lasted. Conclusions 8-Br-cAMP can inhibit HXO-Rb44 cell proliferation by inducing G1 block of cells.
