摘要
目的 :观察吴茱萸氯仿提取物 (主要成分为吴茱萸碱和吴茱萸次碱 )对小鼠实验性结肠炎的作用。方法 :TNBS/乙醇两次灌肠制作小鼠溃疡性结肠炎模型 ,小鼠经口给予吴茱萸氯仿提取物 ( 5 0 ,10 0 ,3 0 0mg/kg× 14d) ,考察吴茱萸的药效作用 ,同时考察吴茱萸对炎症反应中PGE含量的影响。结果 :TNBS诱发结肠炎后 ,小鼠腹泻、便血、结肠出现了粘连、充血 ,溃疡出血甚至穿孔等变化。吴茱萸能改善结肠炎小鼠大体形态学损伤 ,降低MPO含量 ,抑制脾肿大 ,对小鼠一般状况如腹泻、便血、体重变化都有所改善。吴茱萸降低小鼠角叉菜胶致炎足PGE含量。结论 :吴茱萸氯仿提取物能抑制实验性结肠炎模型小鼠的炎性浸润、渗出和增生 ,减轻结肠局部的病理损害。
Objective: To investigate the anti-inflammatory activity of chloroform extract of Evodia rutaecarpa (EVO) on the acute and chronic stages of a trinitrobenzene sulphonic acid (TNBS) experimental model of inflammatory bowel disease (IBD) in mice. Methods: A mouse model of IBD was induced by colonic instillation of TNBS/50% alcohol 0.1ml at 7day intervals (2mg for initial, 1mg for second treatment). EVO 30, 100, 300mg/kg were administrated orally for 14 days. Macroscopic damage score, myeloperoxidase (MPO) activity, colon weight, spleen index, and body weight were determined to access the severity of IBD. The effect of EVO on the activity of Prostaglandin E 2 (PGE 2) in inflammation was also examined. Results: EVO lowered the macroscopic damage score, myeloperoxidase activity, colon weight, spleen index in mice treated with TNBS. And it alleviate the manifestation of colitis such as diarrheal, occult blood in the feces, the decrease of body weight. EVO also lowered the Prostaglandin E 2 in hind paw edema induced by carrageenin in mice. Conclusion: EVO has the anti-inflammatory activity on TNBS experimental model of inflammatory bowel disease (IBD) in mice. That it lowered the PGE 2 perhaps in one of the mechanisms of anti-inflammatory effects of EVO.
