随机选取 1日龄高能日粮饲养的AA肉鸡 ,其中 10 0羽在正常温度 (2 0℃ )下饲养设为对照组 ,15 0羽在低温 (11℃ )环境下饲养设为试验Ⅰ组 ,10 0羽为低温 (11℃ )环境下日粮添加 1%L 精氨酸设为试验Ⅱ组。低温诱发肉鸡腹水综合征 ,分别于 3、4、5、6和 7周龄采集血液 ,检测环核苷酸 (cAMP、cGMP)与血管内皮衍生因子 (ET 1、AngⅡ ) ,观察NO前体物L 精氨酸对AngⅡ的抑制作用及对内皮细胞的保护作用。结果表明 :试验Ⅰ组复制肉鸡腹水综合征的发病率为 9.3% ,试验Ⅱ组及对照组均为 3% ,与试验Ⅰ组相比P <0 .0 1。试验Ⅰ组肉鸡血浆cAMP、cGMP、cAMP/cGMP含量持续升高 ,与相应时间的对照组比较P <0 .0 1。AngⅡ含量逐渐降低 ,但始终高于相应时间的对照组P <0 .0 5 ,ET 1在腹水形成之前高于对照组P <0 .0 5 ,腹水形成后 2者水平无统计学意义P >0 .0 5。发生腹水综合征的肉鸡cAMP、cGMP、AngⅡ、ET 1含量明显高于相应时间的对照组P <0 .0 1、P <0 .0 5。试验Ⅱ组血浆cAMP先升高 ,后降低 ,但仍然高于相应时间的对照组P <0 .0 1,cGMP始终比试验Ⅰ组及对照组高P <0 .0 1,P <0 .0 5 ,cAMP/cGMP和AngⅡ含量随试验时间的延长而逐渐降低 ,与相应时间的对照组比较P <0 .0 5和P <0 .0 1,ET 1变化不大。
A flock of AA breed chickens were reared in peterstme brood vait chamber and fed with energy pelleted feed, at 14 days of age, 400 birds were devided into 3 groups randomly as follows: 100 birds were exposed to normal ambient temperature (20℃, control group),150 birds were exposed to low ambient temperature (11℃) in order to induce ascites (groupⅠ), another 100 birds were also exposed to low ambient temperature (11℃) and fed with diets containing 1% L arginine for prophylactic treatment of ascites (groupⅡ). The blood samples were collected at 3rd, 4th, 5th, 6th, 7th week, respectively, to measure the contents of plasma endothelin (ET 1), angiotensin Ⅱ(Ang Ⅱ), cyclic adenosine monophosphate (cAMP), cyclic guanosine monophosphate (cGMP). The results indicated that the morbidity of ascites in broilers was 9.3% in groupⅠ,while the other two groups were 3%. The contents of cAMP, cGMP, Ang Ⅱ and the ratio of cAMP/cGMP in groupⅠand ascitic broilers were higher than the control group ( P <0.01, P <0.05), ET 1 of preascitic broilers were higher than control group ( P <0.05) , while no significant difference existed in them among ascitic broilers. The contents of cAMP and cGMP in groupⅡ were higher than groupⅠand control group ( P <0.01, P <0.05 ), whereas, the ratio of cAMP/cGMP and the contents of AngⅡwere gradually decreased,but higher than control group( P <0.05). The change of contents of ET 1 were not significant. The increased plasma AngⅡin broilers at low ambient temperature condition made the secretion of ET 1, cAMP, cGMP by endothelium cells increased and the production of NO decreased. So low temperature accelarated ascites syndrome in broilers. Supplementation of L arginine decreased ET 1, cAMP and cAMP/cGMP. It is concluted that cAMP mediated, AngⅡand ET 1 induced pulmonary hypertension syndrome in broilers.
