摘要
目的 :研究低氧 /复氧 (H/R)对心肌细胞内Ca2 + 浓度 ([Ca2 + ]i)的影响以及吡那地尔 (Pinacidil)减轻H/R过程中钙超载的作用。 方法 :采用新生SD大鼠进行心肌细胞培养 ,建立心肌细胞H/R模型。实验分 :①正常对照组 ;②H/R组 :细胞低氧 30min/复氧 30min ;③吡那地尔组 :先加入终浓度为 2 5 μmol/L的吡那地尔 ,再行低氧 30min/复氧 30min。以Fluo 3/AM荧光指示剂负载 ,应用激光共聚焦显微镜技术检测心肌细胞 [Ca2 + ]i变化。 结果 :对照组心肌细胞 [Ca2 + ]i荧光强度和荧光光密度值较低。低氧 30min后复氧即刻 ,[Ca2 + ]i荧光光密度值开始增加 ,复氧 30min后 [Ca2 + ]i荧光强度和荧光光密度值显著增高 (与对照组相比 ,P <0 .0 1 )。而吡那地尔组细胞内荧光光密度值较H/R组显著降低 (P <0 .0 1 )。 结论 :心肌细胞H/R导致钙超载 ;而吡那地尔有明显减轻心肌细胞H/R时Ca2 +
Objectives: To examine effects of pinacidil on intracellular free calcium concentration of cardiomyocytes during hypoxia/reoxygenation. Methods:A cell culture model of neonatal rat cardiac myocytes was used. There were three groups, including control group, hypoxia/reoxygenation group and pinacidil group. Confocal microscope was used with Fluo 3/AM as calcium indicator to detect changes of intracellular free calcium concentration. Results:The intracellular fluoresence intensity of singular cardiomyocyte in hypoxia/reoxygenation group was significantly higher than that of the controls( P <0.01),whereas with pinacidil,the fluoresence intensity was significantly lower than that of the hypoxia/reoxygenation group( P <0.01). Conclusions: Pinacidil decreases intracellular free calcium concentration of cultured neonatal rat cardiomyocytes during hypoxia/reoxygenation.
