摘要
目的 :用反相高效液相色谱法测定陕西不同地区鬼灯檠中岩白菜素的含量。方法 :采用SymmetryC18色谱柱(5 μm ,4 .6mm× 15 0mm) ,流动相为甲醇 水 (2 5∶75 ) ,流速为 1.0mL·min-1,柱温为 (30± 1)°C ,检测波长为 2 75nm。结果 :岩白菜素在 0 .8~ 4 .0 μg呈良好的线性关系 (r =0 .9993) ,方法回收率为 99.95 % ,岩白菜素的相对标准偏差为 3.0 % (n=5 ) ,最低检测限为 0 .1ng。 结论 :本方法快速、简单、灵敏度高 ,岩白菜素含量与原料产地有一定的关系。
Objective: To establish a RP-HPLC for determination of bergenin in Rodgeris aesculifolia batal from different sources. Methods : Chromatographic conditions were as follows: C 18 column(5μm, 4.6mm×150mm,i.d), was adopted, mobile phase consisted of 25% ethanol and 75% water. The column temperature was set at (30±1)°C. The flow rate was 1.0mL·min -1 and the detection wavelength was at 275nm. Results : The method was proved to be linear in the range of 0.8~4.0μg with a regression coefficient of 0.9993. The average recovery was 99.95% with RSD 3.0%( n =5). The minimum detection limit was 0.1ng. Conclusion : The method is proved to be quick, simple and highly sensitive. There is some relationship between the bergenin contant and different sources of raw materials.
