摘要
为了对犬钩虫(Ancylostoma caninum)卵细胞有丝分裂过程中染色体进行观察,首先以0.001%的秋水仙素抑制卵裂时纺锤体的形成,然后用1%的漂白粉溶液脱去卵壳的几丁质层、空气干燥法制片,Giemsa 染色。对35个中期卵染色体数目进行观察,结果发现57%的卵细胞具有12条染色体,26%的卵细胞有11条,初步认为犬钩虫卵细胞染色体为11~12条。
The studies were carried out on chromosome of Ancylostoma caninum in their cleavage divi- sions.The eggs which had been cultivated in feces containing 0.001% colchicinc solution were treated with 1% bleach powder solution.The chitin layer of eggs can be ereased.Followed by drying preparation and staining with 10% Giemsa solution,the chromosome samples had been gotten.35 cells with metaphase from 400 eggs had been studied carefully,20 cells(57%)had the chromosome number 2n=12,9 cells(26%)had the chromosome 2n=11.The somatic chromo- some number in Ancylostoma caninum were either 2n=11 or 2n=12.
