毛头鬼伞CC-8810深层培养工艺的研究

摘要 为了提高发酵终了时毛头鬼伞(Coprinus comatus Fr.)CC-8810的菌丝浓度以获取高等真菌多糖,对CC-8810的深层培养工艺进行了研究。确定了最适发酵条件为:4%葡萄糖、3%黄豆饼粉、0.5%酵母浸出粉、3%麸皮、0.1%KH_2PO_4、0.05%MgSO_4·7H_2O,pH5.5—6,温度28℃,摇床转速140—160 r/min,摇瓶种子培养4天接种。当发酵液pH降至5.0—4.9、残糖量在1.2%以下,5—6天可终止发酵。发酵罐中放大试验表明,最终菌丝体浓度达45.1g/L(干重),含纯多糖3.6g/L。 Suitable conditions for polysaccharide production in submerged culture by Coprinus comatus Ft. CC-8810 were: glucose 40g, Soybean cake powder 30g, yeast extract powder 5.0g, bran 30g, KH_2PO_4 1.0g, MgSO_4·7H_2O 0.5g, tap water 1L, pH5—6, 28℃, 160r/min. Seed culture grown in shake flask for 4 days were inoculated into 1.5L fermentor. After 5—6 days the fermentation ended up with decreases in pH to 4.9—5.0 and in reducing sugar to 1.2%. 45.1g(dry Wt.)/L hyphae and 3.6g/L polysaccharide were obtained.
