研究了大豆豆粕中提取异黄酮的影响因素。考察了乙醇浓度、提取次数、提取时间和溶剂原料比四因素对提取的影响。结果表明,适宜的工艺条件是:70%乙醇,回流提取3次,每次3h,溶剂原料比3∶1,干膏得率为32.73%,异黄酮含量22.24%。研究了干膏中酸解异黄酮的影响因素。考察了盐酸浓度、酸解时间、酸解温度和盐酸体积原料比四因素对酸解的影响。结果表明,适宜的工艺条件是:0.1mol/L盐酸.酸解3h,温度40℃,盐酸体积原料比5∶1,金雀异黄素含量5.95%,分解率为5.25%。
The factors effected on the isoflavone extraction from soybean cake are studied in this paper. The effects of alcohol concentration, times of extraction, ratio of solvent and sample were investigated. The results showed that the optimized conditions were the alcohol concentration 70%, reflux extracting 3 times, 3 hours per times, the ratio of solvent and sample 3:1. The yield of extraction was 32.73%, isoflavone content was 22.4%. The factors effected on the acidolysis of isoflavone in dry paste were also studied. The effects of hydrochloric acid concentration, time, temperature, the ratio of hydrochloric acid and sample on the isoflavone acidolysis were investigated. The results showed that the optimized conditions were 0.1mol/L hydrochloric acid, 3 hours, temperature 40℃, the ratio of hydrochloric acid and sample 5:1. Genistein content 5.95%, the ratio of acidolysis 5.25%.
