应用组织工程方法体外培养人牙囊细胞与I型胶原的合成(英) 被引量:1

摘要 目的:研究体外培养人牙囊细胞,探讨其生物学特性及其在牙齿萌出中的作用。方法:取年龄为12岁人埋藏阻生第三磨牙的牙囊,组织块法进行人牙囊细胞的培养,HE染色、扫描电镜观察人牙囊细胞的形态,免疫组织化学染色技术观察Ⅰ型胶原及波形蛋白在牙囊细胞中的表达及定位。结果:培养的人牙囊细胞呈梭形,卵圆形及多角形,形似成纤维细胞;扫描电镜可见细胞多呈梭形,也可见多角形,所有细胞表面均有颗粒状物质,有的细胞表面多且密,有的细胞表面较少,折光性较强,细胞核周围分布较多;在牙囊细胞中Ⅰ型胶原和波形蛋白的表达呈阳性,定位于细胞的胞质中,围绕细胞核周围染色较强,细胞核内染色阴性。结论:体外可以培养人牙囊细胞,人牙囊细胞具有成纤维细胞的特性,具有合成Ⅰ型胶原的能力。 AIM:The dental follicle is important for the eruption of teeth.The objective of this study is to culture the human dental follicle cells in vitro and research the characterization of these cells to investigate the effects of dental follicle in the teeth eruption. METHODS: The dental follicles were dissected from the lower third impacted molars (uneruption), and then cultured in a medium. The cultured cells grew to confluence and were through 3 passages before experimentation. They were observed with microscopy and scanning electron microscopy. Immunohistochemical staining technique and immunofluorescence staining showed the expression of vimentin and type Ιcollagen respectively in cultured human dental follicle cells and localization of them in cytoplasm. RESULTS: Dental follicle cells cultured were elongate shape, exhibited fibroblastic characteristics, and scanning electron microscopy revealed that there were a lot of granules in the surface of cultured cells.The expression of type Ιcollagen and vimentin showed positive; the localization of type Ιcollagen and vimentin were in the cytoplasm surrounding of nucleus. CONCLUSION: The results indicate that the human dental follicle cells can be cultured in vitro, exhibit fibroblastic characteristics, derive from intermediate filaments, and synthesize type Ιcollagen.
