摘要
目的 利用体外培养大鼠传代星状细胞为模型 ,观察丹参对星状细胞凋亡相关蛋白FasL、Fas、bax和bcl 2表达的调节作用 ,以进一步阐明丹参诱导星状细胞凋亡的机理。方法 大鼠星状细胞的分离采用胶原酶原位循环灌流法。星状细胞经2 0g/L丹参作用 4 8h后 ,用免疫细胞化学法检测FasL、Fas、bax和bcl 2 ,用Westernblot法检测bcl 2表达。结果 免疫细胞化学结果表明 :星状细胞经丹参作用后FasL表达明显增强 ,Fas稍增加 ,bax明显增强 ,bcl 2减少 ;用Westernblot法未能检出bcl 2。结论 丹参通过Fas途径及增加bax表达和减少bcl 2诱导肝星状细胞凋亡。
Objective To investigate the effects of salvia miltiorrhiza on Fas ligand(FasL),Fas,bcl 2 and bax expressed by rat hepatic stellate cells(HSCs) in vitro.Methods HSCs were isolated from rats with collagenase in situ liver recirculating perfusion.Salvia miltiorrhiza(20g/L)was added and incubated for 48 hours.FasL,Fas,bcl 2 and bax were detected by immunohistochimical staining and bcl 2 was detected by Western blot.Results FasL and bax expression were increased in HSCs treated by salvia miltiorrhiza,Fas staining was slightly enhenced.Compared to the control group,bcl 2 was weak in immunostaining and could not be detected in Western blot.Conclusion Apoptosis of rat hepatic stellate cells induced by salvia miltiorrhiza might be modulated through Fas apoptotic pathway with increased bax and decreased bcl 2 expression.
