摘要
目的 测定丹参中丹酚酸B ,丹参素和原儿茶醛等 3种水溶性有效成分的含量。方法 采用高效液相色谱法测定。以甲醇 5 %冰醋酸为流动相进行梯度洗脱 ,以对羟基苯甲酸为内标 ,检测波长为 2 81nm ,柱温 30℃。结果 丹参素、原儿茶醛和丹酚酸B浓度分别在 3.95~ 4 7.4 μg·mL-1(r =0 .9996 ) ,5 .14~ 6 1.6 8μg·mL-1(r =0 .9999) ,16 .8~ 117.6 μg·mL-1(r =0 .9994 )范围内呈良好线性关系 ,丹参素、原儿茶醛和丹酚酸B的平均回收率分别为 10 1.4 3% (RSD =2 .88% ,n =6 ) ,99 .89% (RSD =3.0 4 % ,n =6 )和 10 3.4 3% (RSD =1.5 2 % ,n =6 )。结论 方法简便 ,分离效果好 ,能同时测定丹参中丹酚酸B、丹参素、原儿茶醛等 3种水溶性有效成分的含量 。
OBJECTIVE: To investigate the determination of three water soluble components in Salvia miltiorrhiza. METHODS: The analysis was carried on a column of Hypersil C18(250 mm × 4.6 mm, 5 μm) with a mobile phase of methanol-5% acetic acid (gradient elution from 10:90 to 35:65 in 20 minutes), using 4-hydroxy-benzoic acid as the internal standard. The detection wave-length was at 281 nm. RESULTS: The linearity was obtained over 3.95 ∼ 47.4 μg·mL-1(r=0.999 6) for Danshensu, 5.14 ∼ 61.68 μg·mL-1(r=0.999 9) for protocatechuic aldrhyde and 16.8 ∼ 117.6 μg·mL-1(r=0.999 4) for salviamolic acid B, respectively. The average recoveries were 101.43% (RSD=2.88%, n=6) for Danshensu, 99.89% (RSD=3.04%, n=6) for protocatechuic aldrhyde and 103.43% (RSD=1.52%, n=6) for salviamolic acid B, respectively. CONCLUSION: This method appeared to be rapid, simple and accurate.
