摘要
以不同辐射剂量的 6 0 Co- γ射线处理菊花愈伤组织 ,获得变异后代 ,从分子水平上检测不同辐射剂量对植物遗传物质的影响 ,运用随机扩增多态性 DNA(RAPD)技术对 5种变异株和对照进行分子标记和遗传突变的研究 .建立了适合 RAPD的反应体系 ,优化其反应条件 ,从 2 5种引物中筛选出的 6种引物表现出明显的多态性差异 ,共扩增出 72条带 .结果表明 ,经过不同辐射剂量处理获得的变异菊花在 DNA水平上存在多态性差异 .分析了多态性产生的机理和变异株间遗传物质的差异 ,并讨论了 RAPD的可靠性和辐射变异的机理 .
After^(60)Co-γ ray radiating to the Chrysanthemum callus in different dosage, the offsprings have been changed in many characters. Five varieties and contradistinguish of the radiated Chrysanthemum offspring are adopted as the experimental materials in this experiment,with the application of random amplified polymorphic DNA (RAPD) technology to the molecular genetic marker.Many factors of RAPD procedure may influence the result. This paper presents selected reaction condition for analysis of genetic relationship among them. It is constructed that the systemic technologies of RAPD for the identification of specificity of the Chrysanthemum.As a result, with the 6 materials,6 polymorphism primers are selected by this system from 25 arbitrary primers,and 72 amplified bands are obtained,which can be successfully used identifying the Chrysanthemum varieties and lines.Compared with contradistinguish, the differences of the five varieties have beenfound in the molecular level. According to the analysis of RAPD and previous research, the author analyses the difference of the genetic material, and discusses the mechanism of the variation.
