HIV侵入抑制剂的研究进展

摘要 早期HIV研究的重大突破之一就是发现其主要的细胞受体为CD4分子。而1996年第二受体(second receptor,亦称共受体,coreceptor)的发现和近年来在HIV-1包膜糖蛋白研究方面取得的一系列进展,使人们对HIV入侵机制的了解日益深入,明确了HIV对宿主细胞的入侵有3个关键的步骤:①HIV包膜糖蛋白gp120与宿主细胞上的CD4受体结合;②gp120进一步与第二受体CCR5或CXCR4结合;③gp41暴露,病毒与细胞膜发生融合。在此基础上,一类新的抗HIV药物——侵入抑制剂(entry inhibitor)应运而生,根据每一步不同的靶位点,它们相应分为:粘附抑制剂(attach-ment inhibitor)、受体抑制剂(coreceptor inhibitor)和融合抑制剂(fusion inhibitor)3种,综述如下。 1
