摘要
以高山红景天种子、试管苗的叶片、茎等为外植体,研究了影响愈伤组织诱导和生长的激素浓度和光温条件.结果表明,在浓度为2.0,8.0mg/L的NAA和浓度为3.0,5.0,8.0mg/L的6-BA配合的MS培养基上得到了较好的愈伤组织诱导效果,附加2,4-D时浓度为1.5mg/L的愈伤组织诱导率显著高于0.5mg/L浓度,特别是与浓度为5.0,8.0mg/L的6-BA配合时效果更明显.适合愈伤组织生长的温度为20~24℃,光照长度为12~14h.
The hormone concentration illumination and temperature effecting the introdution and the growth of Rhodiola sachlinensis A.Bor were researched and the seeds of Rhodiola sachlinensis A.Bor, the leaves and stems of test-tube seedling were used as explant. The result indicated that the optimum introduction effect of callus could be achieved by the MS substratum with the coordination of 2.0,8.0 mg/L NAA and 3.0,5.0,8.0 mg/L 6-BA. The callus induction rate of Rhodiola sachlinensis A.Bor was higher if the 1.5 mg/L 2,4-D was added than that of adding 0.5 mg/L 2,4-D, especially coordinating with the 5.0,8.0 mg/L 6-BA. The suitable temperature and illumination time of Rhodiola sachlinensis A.Bor callus was 20~24℃, 12~14 h.
