摘要
应用乳酸脱氢酶释放法对重组白介素2,粗制天然白介素2,重组肿瘤坏死因子和天然免疫活性肽等淋巴因子诱导人PBMNC为LAK细胞以及LAK细胞对靶细胞(人外周血淋巴细胞,人食管癌109细胞株和人红白血病细胞株_(562)的细胞毒作用分别于培养的第4d和第7d进行了检测。结果显示:①四个配伍组和一个重组白介素2组的多克隆LAK细胞攻击溶解109细胞株和K_(562)细胞株的水平波动子28%和66%之间,中位数为47%,而对正常人外周血淋巴细胞均无细胞毒作用;②重组白介素2和粗制天然白介素2配伍培养LAK细胞的增殖协力约高于重组白介素2和重组肿瘤坏死因子组,以及重组白介素2和免疫活性肽组增殖协力的3倍,③在诱导LAK细胞的过程中其增殖量和其细胞毒活性可以不同步。建议临床上治疗晚期肿瘤病人除用LAK细胞和重组白介素2外,首先选用天然白介素2。
In this paper,rIL- 2, impure nlL- 2, rTNF and IAP were used to induce
transforming of PBMNC cultured in RPMI media into LAK cells and the cytotoxicity or LAK cells
against target cells(human allogeneic peripheral blood lymphocyte-PBL, human esophageal
cancer 109 cell line-EC109 and human erythroleukemia cell line- K_(562) was measured by a
LDH release assay at day 4 and 7.The results showed that(1) the polyclonal LAK cells in each
of 4 co-culture media and one medium with rIL-2 alone lysed EC109 and K_(562) at levels
varied between 28%and 66% ith a medi-an of 47%but possessed no the activity of lysing
normal human periph-eral blood lymphocytes;(2)the synergism in proliferating of LAK cells in
medium supplemented with rIL-2 and impure nIL-2 was approximately 3 times that caused by
the additive effect of rIL-2 with either rTNF or IAP;(3)the proliferation and cytotoxicity of LAK cells
might not be synchro-nous in the induction course.It is therefore suggested from the data
sum-marized above that the impure nIL-2 be selected clinically and preferentially, in addition
to LAK cells and rIL-2,to cure the patients with advanced cancer.
