摘要
为了评估脐血中间充质干 /祖细胞存在的频率 ,了解脐血间充质干 /祖细胞的增殖特点 ,寻找新的组织工程种子细胞 ,取足月顺产胎儿脐血、淋巴细胞分离液分离单个核细胞 ,悬浮至含 2 %胎牛血清的DMEM培养基中 ,培养并扩增脐血间充质干 /祖细胞 ,行免疫组织化学鉴定。结果表明 :脐血单个核细胞中间充质干 /祖细胞集落出现的频率为 0 .5× 10 -6,呈成纤维细胞样 ,传 2 0代仍可维持其形态特征 ,可有效扩增 1.3× 10 7倍。结论 :低血清DMEM培养液能维持脐血间充质干 /祖细胞增殖分化能力 ,脐血间充质干 /祖细胞是又一良好的组织工程与细胞治疗的种子细胞。
This study was done for investigating the frequency and proliferative feature of mesenchymal stem/progenitor cells (MSPC) in human umbilical cord blood (CB) and for searching a new seed cell for tissue engineering. Mononuclear cells was separated by Ficoll Hypaque from cord blood and suspended in DMEM culture medium supplemented by 2% fetal bovine serum. The adherent CB cells were cultured and expanded at same medium. The results showed that the frequency of CB MSPC was 0.5×10 -6 [(0.2-0.8)×10 -6 ]. The CB MSPC showed a fibroblast like morphology and retained their morphological feature at least after 20 sub passages, and could extensively be expanded by about 1.3×10 7 times as much. The conclusion is that low serum DMEM culture could maintain the proliferation and differentiation potential of CB MSPC. CB MSPC might be a favorable seed cell for tissue engineering and regeneration.
