摘要
目的将基因芯片技术用于鼠疫的快速诊断。方法分别采用RD标记技术对鼠疫耶尔森菌、小肠结肠炎耶尔森菌和假结核耶尔森菌的DNA样品进行标记,与芯片上的探针进行杂交。结果芯片能将鼠疫耶尔森菌与同属的2种致病性细菌鉴别开。结论基因芯片是一种有效的诊断工具,可用于鼠疫的诊断。
Objective To develop a DNA microarray t echnique for fast diagnosis of plague. Methods Restriction display polymerase chain reaction (RD-PCR) and hybridization with fluorescently labeled cy5 were em ployed for detecting the sam-ple DNA of Yersinia pestis, Yersinia pseudotubercul osis and Yersinia entercolitica. Results The prepared microarray is capa-ble of distinguishing Yersinia pestis from Yersinia pseudotuberculosis and Yersinia ent ercolitica in the same genus. Conclu-sion The constructed DNA microarray is an effective diagnostic tool for the detection of Yersinia pestis.
