摘要
目的 探讨血管增生在CML(慢性髓细胞性白血病 )病程中的变化 ,为CML抗血管新生药物治疗提供实验依据。方法 应用ELISA法检测CML患者与正常人群血浆中VEGF、TXB2、GMP 14 0的水平 ,并观察CML患者血浆作用下内皮细胞株在纤维蛋白、Ⅳ型胶原上迁移能力的改变。结果 CML患者血浆中VEGF、TXB2、GMP 14 0水平较正常对照显著增高 ,其中治疗前水平又较治疗后高。CML患者血浆作用下内皮细胞株迁移能力增强 ,以治疗前为甚。结论 CML患者存在血管增生 ,病情缓解后 ,血管增生也得到了缓解。提示不仅血管增生检测指标可以作为CML等恶性肿瘤疗效及预后判断参数 。
Aim To study the variation of angiogenesis in the developmen t of chronic myeloid leukemia (CML) and provide the experimental data for the an ti-angiogenesis drugs in the application of the therapy to CML.Methods ELISA was used to detect the plasma level of VEGF, TXB2 and GMP-140 i n the normal control and patients with CML and to observe, the mobility of endot helial cell line (ECV304) on the surface of fibrinogen and type Ⅳ collagen. Results Compared with normal control, the plasma level of VEGF, TX B2 and GMP-140 in the patients with CML increased and additionally the plasma l evel of the three parameters were higher in the CML patients without therapy tha n those after alleviation. The mobility of ECV304 on the surface of fibrinogen a nd type Ⅳ collagen was enhanced under the effect of plasma from CML patients an d this phenomenon was more obvious in the patients without therapy. Conc lusion This study showed that there were variations of angiogenesis in the development of CML. The parameters about the angiogenesis were not only eva luative for the judgment of curative effect but also for judgment of the prognos is of malignant tumors. Anti-angiogenesis method had become a novel potent path way in the therapy of malignant tumors.
