鸡卵黄抗眼镜王蛇毒抗体IgY的理化特性 被引量:2

The Study on Physico-chemical Characterization of Anti-king Cobra Venom's IgY from Hen Egg Yolk

摘要 目的 研制抗眼镜王蛇毒鸡卵黄抗体并研究该抗体的理化特性。 方法 拟用减毒后的眼镜王蛇毒免疫母鸡后 ,从卵黄中制备抗眼镜王蛇毒抗体 Ig Y。采用间接 ELISA法对此抗体的理化特性进行研究。 结果  ELISA显示免疫后 12天开始出现特异性抗体 ,且抗体滴度逐渐升高 ,约在免疫后 6 0天左右 ,最高可达 1:10 0 0 0 0 ,此最高滴度可维持 30天 ,以后滴度逐渐降低 ,至免疫后 10 0~ 110天 ,滴度仍可维持在最高滴度的一半 ( 1:50 0 0 0 )。此抗体在 10~ 6 5℃温度范围内活性保持稳定 ;在 p H为 4~ 12范围内 ,抗体活性稳定 ;此抗体经胰蛋白酶处理 1h内 ,活性下降不明显 ,但 1h以后活性迅速下降。 结论 从卵黄中制备抗眼镜王蛇毒抗体 Ig Y,是可行的 ,稳定的。 Objective To explore the production of anti king cobro venom's antibody from hen egg yolk and this antibody's charcteristic effect Methods We use king cobra,it's venom has been decreased, as our antigen to immune hen By Enzyme linked immunoadsorption assay( ELISA),this experiment study charcteristic effect of IgY which is the antibody the hen producing and transfering to egg yolk Results ELISA demonstrates the antibody was first appeared after immunized 12 days and then titer raised gradually ,reach to 1:100000, the highest titer (about after immunized 60 days ), then the titer decreased,after immunized 100~110 days ,the antibody's titer also can maintain at 1:50000 IgY has stable activity It's stability can maintain between 10~ 65℃; IgY can keep it's activity between pH value 4 to 12 when test in 37℃ 2 hours; When the IgY is mixed with trypsin, it's activity change little in one hour, but miss quickly after 2 hours Conclusion The anti king cobro antibody that have been produced has stable activity So that it is available to apply this method to produce antibodys
