重叠区扩增基因拼接法拼接hIL-2信号肽基因和CVB3的VP1基因

SPLICING HUMAN IL-2 SIGNAL DNA SEQUENCE AND CVB3 VP1 GENE BY OVERLAP EXTENTION

摘要 目的 :将 h IL- 2信号肽基因拼接到 CVB3VP1基因的 5′端 ,以获得分泌型 VP1 (s VP1 )基因。方法 :采用重叠区扩增基因拼接法。结果 :凝胶电泳见到预期大小 DNA条带 ,经测序表明基因序列与报导的 h IL- 2信号肽及 CVB3VP1的基因序列一致。结论 :成功获得了融合基因 s Objective:To construct secretable VP1(sVP1) gene by fusing the signal DNA sequence of hIL 2 to the 5′terminal of VP1 gene.Methods: Genes were spliced by overlap extension.Results: Agarose gel electrophoresis analysis showed the expected DNA bands. DNA sequencing showed that the sequence of the sVP1 cDNA was identical with that reported for VP1 cDNA and hIL 2 signal sequence.Conclusions: The signal DNA sequence of hIL 2 was fused to the 5′terminal of VP1 gene and sVP1 cDNA was constructed successfully.
