摘要
目的 :观察舒心胶囊对缺氧导致人脐静脉内皮细胞 (HUVECs)损伤的保护作用。方法 :体外培养HUVECs,建立缺氧模型 ,以透射电子显微镜技术观察细胞的微观结构 ;检测细胞培养液中LDH、NO、ET - 1、Ang -Ⅱ及 6 -keto -PGF1α水平。结果 :舒心胶囊能够稳定HUVECs的微观结构 ,显著降低细胞内LDH的释放及ET - 1的分泌 (P <0 0 1) ,增加NO、PGI2 的分泌 (P <0 0 1)。结论 :舒心胶囊对缺氧导致HUVECs损伤具有明显的保护作用。
Objective:To investigate the protective effect of shuxin capsule on cultured human umbilical vein endothelial cells(HUVECs) injured by hypoxia.Methods :HUVECs were cultured in vitro and hypoxic models were established,fine structure of HUVECs were observed by electron microscope,levels of lactate dehydrogenase(LDH)、nitric oxide(NO)、endothelin(ET)、angiotension-Ⅱ(Ang-Ⅱ) and 6-keto-prostaglandin F 1α (6-keto-PGF 1α ) in cultured were examined. Results: Shuxin capsule could notably lessen the damage of HUVECs induced by hypoxia,obviously decrease the release of LDH and secretion of ET-1 ( P <0 01);at the same time, shuxin capsule could also significantly increase secretion of NO and PGI 2 (P <0 01).Conclusion:It is suggested that shuxin capsule can protect HUVECs from hypoxia-injury.
