丙泊酚对大鼠小肠缺血-再灌注后肺损伤氧自由基变化的影响 被引量:3

摘要 目的 探讨不同临床剂量丙泊酚对大鼠小肠缺血-再灌注后肺损伤氧自由基变化的影响及其保护作用。方法 雄性SD大鼠40只,随机分为假手术对照组(C组)、小肠缺血-再灌注组(R组)及分别给予丙泊酚4mg·kg^(-1)·h^(-1)、8mg·kg^(-1)·h^(-1)、10mg·kg^(-1)·h^(-1)(P4、P8、P10)五组;制作小肠缺血-再灌注模型。实验结束即刻留取血样和肺组织,待测脂质过氧化物和肺组织形态学变化。结果 R、P4组血中脂质过氧化物明显升高,与C、P8、P10组相比均有非常显著差异(P<0.01),P8、P10组与C组相比无统计学差异;肺组织脂质过氧化物各实验组与对照组相比差异非常显著(P<0.01),以R组改变最为明显;各用药组指标均得以改善,与R组相比有显著差异(P<0.01),P4与P8、P10组相比亦差异显著(P<0.01)。假手术组肺组织结构正常,R组可见肺泡压闭实变,腔内有大量渗出,炎性细胞浸润,毛细血管扩张,支气管壁增厚,各用药组肺部改变较R组明显减轻,P8、P10两组除了有少量改变外,肺组织结构基本接近正常。结论 氧自由基在大鼠小肠缺血-再灌注后肺损伤中有重要作用,临床剂量丙泊酚不同程度地抑制脂质过氧化反应,对小肠缺血-再灌注后氧自由基介导的肺损伤产生保护作用。 Objective To study the effects of different dose propofol on the changes of reactive oxygen species after intestinal ischemia-reperfusion induced lung injury in rats. Methods Forty male Sprague Dawley rats, weighing 200-250 mg,were randomly divided into 5 grpups of control(subjected to sham operation) ,R(super mesenteric artery occlusion 60 min and reperfusion 120 min) ,P4,P8,P10 (subjected to SMA occlusion,and at 20 min before clip removal perfused continually propofol 4 mg · kg-1· h-1 ,8 mg · kg-1 · h-1 and 10 mg · kg-1 · h-1 respectively. ). Lipid peroxides(LP)of blood and lung tissue were measured, and morphologic changes of the lungs were examined. Results Serum LP concentration of group R and P4 increased obviously after IIR, compared to that of group C,P8 and P10. The histological examination of the lungs revealed negligible alterations in the group P8 and P10, whereas the group IIR showed a marked increase in infiltration of the interstitial space and substantial thickening of the alvolar septa. Conclusions The reactive oxygen species plays an important role for the lung injury induced by intestinal ischemia-reperfusion,and the clinical dose propofol has protective effects on the lung injury by restraining the lipid peroxidation.
