卵巢癌细胞培养上清抑制CD8^+ T细胞增殖及IL-2R表达的研究 被引量:1

摘要 目的 研究不同卵巢癌细胞株培养上清液对CD8+ T细胞增殖功能和表面IL 2Rα、β、γc 链表达的影响 ,探讨卵巢癌腹腔局部免疫抑制的形成机制。方法 将 3种卵巢癌细胞株 (OVCAR3,CAOV3,SKOV3)培养上清液和普通培养液混合培养CD8+ T细胞 ,MTT法测定CD8+ T细胞的增殖反应 ,流式细胞仪检测细胞表面IL 2Rα、β、γc 链的表达 ,ELISA法测定培养上清液中sIL 2R的浓度。结果(1) 3种不同浓度卵巢癌细胞株培养上清液显著抑制CD8+ T细胞的增殖 ,并随浓度增加而增加 ,和对照组比较 ,2 5 %浓度时P值依次为 0 .0 0 4、0 .0 0 6、0 .0 13;5 0 %浓度时P值依次为 0 .0 0 2、0 .0 0 3、0 .0 0 5 ;75 %浓度时P值均为 0 .0 0 1。 (2 ) 3种卵巢癌细胞上清均抑制CD8+ T细胞表面IL 2R表达 ,其中IL 2Rβ和γc 链表达被显著抑制 ,P值依次为 0 .0 0 2、0 .0 13、0 .0 11和 0 .0 15、0 .0 39、0 .0 4 0 ,对IL 2Rα链表达有抑制趋势 ,但差异无显著性 ,P值分别为 0 .2 35、0 .2 2 4、0 .4 98。 (3) 3种卵巢癌细胞株培养上清液均使CD8+ T细胞培养上清中sIL 2R浓度显著上升 ,P值分别为 0 .0 0 8、0 .0 10、0 .0 0 5。结论 卵巢癌细胞株培养上清液对CD8+ T细胞的增殖和IL 2R表达有抑制作用 ,可能是卵巢癌腹腔局部免疫缺陷的机制之一。 Objective To study the effect of supernatants derived from three ovarian carcinoma cell lines on proliferation and function of CD8 + T cell and to investigate the mechanism by which ovarian carcinoma escapes from immune recognition. Methods CD8 + T cells were cultured in complete RPMI 1640 with or without supernatants of three ovarian carcinoma cell lines (OVCAR 3, CAOV 3 and SKOV 3). Proliferation of CD8 + T cell was determined by MTT. Flow cytometry was used to detect the expressions of IL-2Rα, β and γ c on CD8 + T cell surface. ELISA was used to detect the concentration of sIL-2R in the CD8 + T cell supernatants. Results (1) All culture supernatants (OVCAR 3, CAOV 3 and SKOV 3) derived from three ovarian carcinoma cell lines inhibited the proliferation of CD8 + T cell. When the concentration of supernatants was 25%, P value was 0.004,0.006,0.013 respectively; when the concentration of supernatants was 50%, P value was 0.002, 0.003, 0.005 respectively; when the concentration of supernatants was 75%, P value was 0.001. The degree of inhibition was dose dependent. (2) The expressions of IL-2R β and γ c on CD8 + T cell surface were down-regulated significantly by all ovarian carcinoma cell culture supernatants, P value was 0.002, 0.013, 0.011 and 0.015, 0.039, 0.040 respectively. The expression of IL-2Rα was also down-regulated, but not significantly, P value was 0.235, 0.224 and 0.498 respectively. (3) The concentration of sIL-2R significantly elevated in the supernatant of CD8 + T cell which was cultured with ovarian carcinoma cell supernatants, P value was 0.008, 0.010 and 0.005 respectively. Conclusion Soluble factors derived from ovarian carcinoma inhibited the proliferation and function of CD8 + T cell, which might be one of the mechanisms by which the local immunity in the peritoneal cavity of ovarian carcinoma was suppressed.
