Separation and culture of neural stem cells from adult mouse CNS in vitro

摘要 目的 探讨小鼠神经干细胞的体外分离培养方法。方法 采用贴壁培养法,从成体小鼠大脑皮层分离并体外培养成体小鼠大脑皮层细胞,制成细胞悬液后将其置入DMEN/F12(1:1)液(包括15%FCS,bFGF20ng/ml,青霉素100u/ml和链霉素100u/ml)培养,获得细胞克隆。应用免疫组织化学技术检测克隆细胞巢蛋白(Nestin)的表达。结果原代及传代培养的细胞均可持续分裂增殖形成细胞克隆;克隆细胞表达巢蛋白(Nestin)。结论 分离的细胞在体外具有很强的增殖能力和自我更新能力,表达Nestin的为中枢神经系统的干细胞。 Objective To search for a method of the isolation and cultivation of neural stem cells from adult mouse central nervous system (CNS) in vitro. Methods The cerebral cortex tissues were isolated from adult mice. Then suspension of cerebral cortex cells was made. The isolated cerebral cortex cells were cultured in DMEM/F12 (1 :1) solution (consisting of 15%FCS, bFGF20ng/ml, penicilin 100u/ml, and streptomycin 100u/ml). The attachment culture method was used to acquire the clone cells. The expression of nestin antigen of clone were detected by immunohistochemistry technique. Results Cells of primary and passage'cultured continuously proliferated and formed clone cells. The clones expressed nestin antigen. Conclusion Isolated cells which express nestin antigen have abilities to renew themselves, in vitro, so they are stem cells of the CNS.
