摘要
目的 :建立一种较为理想的适用于膜片钳制技术的神经元培养方法 ,并观察其形态学和电生理学特性。方法 :改进的神经元原代培养方法及膜片钳单通道记录法。结果 :采用此方法能获得形态典型的神经元 ,用膜片钳单通道记录法能记录到两类正常的通道电流 (KCa和KATP)。结论 :该培养方法获得细胞数量多 ,细胞质量高 ,电生理特性正常。
Objective: To establish a perfect culture method of cerebral neurons which adapted to patch clamp experiments,and to identifiy the properties of morphology and electrophysiology of the cells.Methods:Cerebral neuron primary culture method was improved.Single channel current was recorded by using patch clamp technique.Results:Using this culture method,cells could be gained which were typical in shape and presented two kinds of channel currents (K Ca and K ATP ).Conclusion:The culture method can produce large numbers of single cerebral neurons which have good quality as well as normal electrophysiologic properties.
