运动心脏重塑中细胞凋亡的基因调控 被引量:14

The Gene Regulation of Cardiomyocyte Apoptosis in Exercise-induced Heart Remodeling

摘要 本研究选用健康成年雄性Sprague-Dawley大鼠 35只 ,随机分为四种不同负荷运动训练组 ,以建立运动性心脏重塑的实验动物模型 ,采用半定量反转录聚合酶链反应技术 ,观察和分析在运动性心脏重塑过程中 ,细胞凋亡调控基因的表达情况及其与运动负荷的关系。结果发现 ,运动训练使心肌组织BaxmRNA表达量增加 ,Bcl- 2mRNA表达量减少 ,Bax/Bcl- 2比值增加 ,Bax与Bcl-2作为调控细胞凋亡发生的中心环节 。 male Sprague-Dauley rats were randomly divided into sedentary group(C), medium-intensity training group (MT), high-intensity training group (HT), once exhaustive exercise group (CE) and once exhaustive exercise after medium-intensity training group (ME). The rats of MT and HT groups ran on treadmill at the densities of 60~80%VO 2max and 80~90%VO 2max for 12weeks, respectively. The expression of apoptosis regulatory genes Bax and Bcl-2 mRNA were tested by RT-PCR.The results showed that the expressions of Bcl-2 mRNA were decreased, the expressions of Bax mRNA and Bax to Bcl-2 ratio were increased in all exercise groups as compared with their controls, respectively. Bax/Bcl-2 ratio in HT and ME groups were significantly higher than that in C and MT groups, respectively. The results suggested that the expression of Bax mRNA and Bax/Bcl-2 ratio had significantly positive correlation with cardiomyocyte apoptosis ratio. The expression of Bax was upregulation, The expression of Bcl-2 was downregulation, The interaction between Bax and Bcl-2 determined the changes and development of cardiomyocyte apoptosis during the exercise-induced cardiac remodeling.
