黄芪在幼鼠肠缺血再灌注损伤下对免疫器官凋亡的作用及意义 被引量:4

摘要 目的 :探讨黄芪在预防幼鼠肠缺血再灌注下 (ischemia/reperfusion ,I/R)免疫器官凋亡的发生的作用及机制。方法 :在幼鼠肠I/R早期 ,电镜观察免疫器官的细胞凋亡 ,DNA原位末端标记法检测细胞凋亡 ,免疫组化法检测Fas/Fas L蛋白的表达。结果 :黄芪在幼鼠肠I/R早期明显抑制脾、肠系膜淋巴结的淋巴细胞、巨噬细胞凋亡。在脾脏 ,明显下调巨噬细胞Fas蛋白的表达 ;在淋巴结 ,明显下调巨噬细胞Fas L蛋白的表达。结论 :黄芪注射液在幼鼠肠I/R早期 ,可能通过影响外周免疫器官Fas/Fas L蛋白表达 ,引发细胞凋亡减少 ,直接影响免疫功能。 Objective: Our aim was to discuss the Astragallus root's protective effects and its mechanisms on immune organs in immature rats of gut ischemia/reperfusion(I/R) model. Methods:Apoptosis were observed with electric microscope and detected by TUNEL in immune organs. The expression of Fas/Fas L was detected by using immunohistochemistry methods in immune organs. Results:Astragallus root significantly decreased apoptosis of lymphocytes and macrophages in the spleen and mesentery lymph node in the earlier period of gut I/R. In spleen, the expression of Fas protein in macrophages was significantly decreased by Astragallus root. In mesentery lymph node, the expression of Fas L protein in macrophages was decreased by Astragallus root. Conclusion:Astragallus root has direct effects on immune state of immature rats in the early stage of I/R, through the affecting on Fas/Fas L expression on secondary immune organ.
