大鼠脑损伤后海马GAP-43 mRNA水平的变化

摘要 以DIG(Digoxigenin)标记的GAP 4 3cDNA片段为探针 ,使用原位杂交方法 ,检测了大鼠皮层硬性脑挫伤后GAP 4 3mRNA水平的变化。结果表明 :海马CA3、CA1和DG区的GAP 4 3mRNA水平在损伤后 4 8h明显升高 ,其中CA3区变化最甚 ,而且伤侧比对侧显著。 1周和 2周后表达仍维持较高水平 ,但不如 4 8h的高 ,这显示表达的峰值在 1周之内。上述结果为研究脑损伤后生理及机能代偿的修复机制提供了有意义的信息。 With DIG (digoxigenin) labeled GAP-43 cDNA as probe, the in Situ hybrid iztion method was set up with the rat hippocampus slides as positive controls. T he change of GAP-43 gene expression after traumatic brain injury was detected a t CA3, CA1 and DG fields of hippocampus. The level of GAP-43 mRNA rised greatly 48?h after brain injury, in which the CA3 field was the greatest, and the inju ried side rised more greatly than the opposite side. The high expression of GAP -43 g ene sustained to 1 and 2 weeks after the operation, but lower than that of 48?h , the period of the highest gene expression is some time in 1 week. The results demonstrate the relationship between brain injury and hippocampus GAP-43 mRNA level, and provide the useful information for the mechanism of pla sticity of cent ral nervous system and physiological functional compensation following traumatic brain injury.
