邻苯三酚红—蛋白质体系的共振光散射光谱研究 被引量:1

Resonance light scattering of protein-pyrogallol red system and its analytical application

摘要 本文研究了邻苯三酚红(PR)与蛋白质的结合反应。在pH3 3的Britton-Robison(BR)缓冲溶液中,PR与蛋白质通过分子间作用力形成缔合物,使最大波长约为385nm的共振光散射光谱得以加强。以PR为标记物根据其共振光散射的增强程度,可用于蛋白质的定量测定。对牛血清白蛋白,测定的线性范围为0-5 0mg L。该方法已用于人尿样品中蛋白质含量的测定。 The binding reactions of the pyrogallol red with proteins have been studied.In Britton-Robison buffer of pH 3.3 the pyrogallol red combines with proteins by intermolecular force to form complexes,causing an enhance of resonance light scattering (RLS)at maximum wavelength of about 385nm.Based on the enhancement of RLS,a novel method for the determination of proteins using pyrogallol red as a labeling agent was developed.The linear range for the determination of proteins is 0 to 5.0mg/L.The method has been used to determine proteins in human urine samples with satisfactory results.
