摘要
目的 :建立一个可用于定量测定全血中 6种氨基酸的纸片采样 高效液相色谱分析方法。方法 :采血样于滤纸上 ,用2 ,4 二硝基氟苯 (DNFB)衍生化 ,HPLC分析测定。结果 :本分析方法在 0 .5~ 10mg·L-1的浓度范围内呈良好线性 ;方法的平均回收率分别为 :缬氨酸 99.0 %、蛋氨酸 90 .5 %、异亮氨酸 95 .1%、亮氨酸 97.4 %、苯丙氨酸 94 .6 %、酪氨酸 96 .6 %。结论 :本方法经济简便 ,适宜于新生儿及儿童的全血中氨基酸浓度测定。
OBJECTIVE To develop a quantitive method with sampling on paper-HPLC for the determination of six amino acids in whole blood.METHODS Small amount of whole blood was absorbed on a piece of filter paper which was derivatized with 2,4-dinitro-1-fluorobezene,and analyzed with HPLC.RESULTS The method was linear within range of 0.5 ~10 mg·L -1 ,and the average recoveries for each of the six amino acids were Val: 99.0% 、Met: 90.5% 、Ile: 95.1% 、Leu: 97.4% 、Phe: 94.6% 、Tye: 96.6% ,respectively.CONCLUSIONS The method is easy to perform for the determination of blood amino acid concentration of the newborn and children.
