摘要
目的 :建立新分离呼肠病毒感染食蟹猴动物模型。方法 :通过滴鼻和静脉感染两种途径 ,用从SARS患者分离的呼肠病毒BYD1株感染 4只食蟹猴 ,每天观察体温变化 ;每组分别在感染后 7d和 33d各处死 1只 ,进行肺脏组织病理学检查和肺脏等脏器的病毒分离。定期采集血清和粪便 ,分别进行特异性中和抗体的测定和病毒分离。结果 :感染猴于接种病毒后的 3~ 4d均开始出现发热症状 ,持续 4d ,体温最高可达 40 .4℃ ,退热 4d后又陆续出现第二次发热。感染动物均产生抗呼肠病毒的中和抗体 ,静脉组抗体滴度高于滴鼻组。从感染动物的肺、心、肝、脾、肾、脑和淋巴组织以及粪便中均分离出呼肠病毒。肺脏组织病理学检查为间质性肺炎。结论 :食蟹猴实验感染呼肠病毒BYD1株后 ,出现发热、免疫应答、病毒血症和间质性肺炎等一系列临床病理学体征 ,说明食蟹猴可以用作呼肠病毒感染实验动物模型。
Objective:To establish an animal model of infection of cynomolgus monkeys(crab-eating monkeys) by a reovirus strain BYD1.Methods:Four cynomolgus monkeys were divided into two groups. Two were inoculated with a reovirus strain BYD1 isolated from SARS patients using injection into the leg vein with a syringe. Two others were inoculated using dropping into the nose. One monkey from each group was sacrificed on day 7 and day 33,respectively. During the infection period, temperatures of the infected monkeys were monitored three times a day. Meanwhile blood and fecal samples were also periodically collected for specific immunology determination and virus isolation. Upon death, monkeys′ lungs were examined for any histopathological changes and their organ samples were collected and cultured for virus isolation. Results:The infected monkeys began to have fever from the third or fourth day after inoculation with the reovirus and it remained for 4-6 days. The highest temperature reached up to 40.4℃. The second attach of fever occurred after their recovery phase. All of infected monkeys produced a specific antibody against this reovirus and the levels of antibody in monkeys infected by inoculation to the leg vein were higher than those through the nose. The interstitial pneumonia was observed in infected monkeys. Reovirus was isolated from the lung, heart, liver, spleen, kidney, brain,lymphatic tissues and feces of infected monkeys by cell cultures.Conclusion:The reovirus strain BYD1 can induce a series of clinical pathological syndrome in cynomolgus monkeys such as fever, immunological responses, viremia and interstitial pneumonia. It is suggested that cynomolgus monkeys can be used as an experimental animal model to study the infection by the reovirus.
