摘要
应用RT-PCR分析转录因子GATA-3基因在慢性粒细胞白血病(CML)中的表达情况。表达GATA-3基因的标本进一步应用单链构象多态性(SSCP)分析GATA-3基因的锌指结构区,发现异常电泳迁移带的标本进行序列分析。结果显示:18/28例CML外用血单个核细胞有GATA-3基因表达,SSCP分析发现一例CML出现异常迁移带,序列分析显示为基因点突变引起精氨酸改变为赖氨酸。结果提示,GATA-3表达于大多数GML中,并存在突变情况。
The expression of transcription factor GAT A-3 gene was detected in chronic myeloid leukemia (CML) samples by reverse RT-PCR. The GATA-3 zinc finger region of all cases with detectable expression was investigated for mutations by single strand conformation polymorphism (SSCP) analysis. Aberrant migrating bands were subsequently sequenced. The result showed that GATA-3 expression was found in peripheral blood mononuclear cells from 18 of 28 CML cases. In one of these 18 cases, bands with aberrant migration were observed upon SSCP analysis. Nucleotide sequencing revealed a point mutation, leading to an amino acid substitution of arginine for lysine. In conclusion, GATA-3 expression is frequently found in CML cell samples and GATA-3 transcripts may display mutation.
