摘要
以RT PCR方法从人工感染禽劳斯肉瘤病毒 (Roussarcomavirus ,RSV )SR RSV的SPF雏鸡体内及SPF鸡胚成纤维细胞 (CEF)培养物中分别扩增出 72 0bp的 gp2 7片段 ,将RT PCR产物重组到质粒载体 pUC19中 ,并对重组质粒进行酶切分析及序列测定。结果表明 ,克隆片段为完整的目的基因 ,该片段的核苷酸序列与国外分离株RSVJ0 2 342的同源性达 97.3%。
The gp27 gene of 720 bp was amplified by RT-PCR method from SPF chicken and Chicken Embyo Fibroblast (CEF) which were infected with Rous sarcoma virus (RSV), and the gp27 gene was cloned into a plasmid veetor pUC19, the recombined plasmid were analyzed by restriction enzyme digestion and sequence analysis, the result showed that the cloning gene was the full-length target gene which has a 97.7% homology with foreign isolate strains RSV(J02342).
