大鼠输精管的神经分布及递质定位的探讨 被引量:2

A STUDY ON THE INNERVATION AND NEUROTRANSMITTERS LOCALIZATION IN THE RAT VAS DEFERENS

摘要 本文应用组织化学、乙醛酸诱发荧光等法显示肾上腺素能(NA)神经;胆碱酯酶(AchE)直接染色法显示胆碱能神经;免疫组织化学 PAP 法显示血管活性肠肽(VIP)与神经肽 Y(NPY)免疫反应神经。对大鼠输精管的观察表明,VIP 免疫反应性神经与胆碱能神经分布基本一致,主要位于固有层内,而 NPY 免疫反应神经与肾上腺素能神经分布基本一致,主要位于输精管肌层。于第1、2、6、7天腹腔注射6-OHDA 4次,第8天处死动物进行观察,此时输精管肌层中的肾上腺素能神经几乎全部消失,其减少程度与 NPY 免疫反应神经基本一致;而 VIP 与胆碱能神经未见变化。免疫电镜 PAP 包埋前染色法显示,VIP 免疫复合物沉积于含直径40~55nm 的清亮小囊泡的膨体中,有的膨体除清亮小囊泡外,还含有少量的大颗粒囊泡(直径100~144nm),NPY 免疫复合物主要沉积于大颗粒囊泡中,偶见于直径40~55nm 的小颗粒囊泡中。以上实验结果提示,在大鼠输精管,VIP 可能与 ACh 共存,NPY 可能与去甲肾上腺素共存。 In this study gloxylic acid-induced fluorescent histochemical method was used to reveal the innervation of adrenergic nerves,acetycholinesterase (AChE) method to show the cholinergic nerves,and the immunohistochemical method to display the vasoactive intestinal peptide (VIP) and neuropeptide Y (NPY) immunoreactive nerves.This observation on the vas deferens showed that the VIP-immunoreactive nerves had the same distribution as the cholinergic nerves,and mainly existed in lamina propria,while NPY-immunoreactive nerves had the same distribution as adrenergic nerves and mainly existed in the muscular layer.One group of animals was treated by intraperitoneal injections of 6-OHDA on the lst,2nd,6th and 7th day,and killed on the 8th day.Following treatment with 6-OHDA,significant evanescence of most adrenergic nerves was observed.NPY-immunoreactive nerves reduced remarkably and the extent of the decrease was similiar to that of adrenergic nerves,whereas no change was observed on the VIP and cholinergic nerves.The immunoelectronic microscopy (PAP pre-embedding method) demonstrated that VIP immunoreactive complex deposited in the varicosity containing small clear vesicles (diameter of 40-55nm).Some varicosities also contained small number of large granular vesicles with the diameter of 100-144mm.NPY immunoreactive complex deposited in the large granular vesicles,and occasionally can be observed in the small granular vesicles,40-55nm in diameter.The results from this study suggest that VIP probably co-exist with ACh and NPY with noradrenalin in rat vas deferens.
