摘要
将老年(26~27个月)及成年(3~4个月)大鼠脑切片用乙醛酸诱发单胺类荧光法,显示其黑质多巴胺(DA)。形态学观察见到,成年大鼠黑质神经元胞体充满强蓝绿色 DA 荧光,突起近端亦显荧光;而老年大鼠黑质神经元胞体蓝绿色 DA 荧光明显减弱,代之以较强的金黄色脂褐素荧光,DA 荧光细胞数量减少。显微荧光光度术对神经元 DA 荧光的定性和定量分析表明,DA 荧光发射光谱峰值为490 nm;脂褐素荧光发射光谱峰值为550 nm。成年组 DA 荧光强度((?)±S_(?))为55.813±3.032,老年组 DA 荧光强度((?)±S_(?))为28.905±2.746,P<0.001,说明老年大鼠黑质DA 浓度较成年组显著降低。本文将所得结果与已有的研究进行了对比分析,并对 DA 降低的可能影响进行了初步讨论。
Age-related changes of dopamine in substantia nigra neurons of adult(3-4
months)and old(26-27 months)Wistar rats were studied by using glyoxylic acid
method to show fluorescence of catecholamine.
There were intensive blue-green fluorescence of DA in the cell bodies and the
proximal part of the processes of the neurons in the adult rats.The fluorescence of
DA in the aging rats was markedly weak and substituted by strong gold-yellow
fluorescence of lipofuscin.The number of DA neurons in the aging rats seemed to
be reduced.
Qualitative and quantitative studies of DA fluorescence in substantia nigra
neurons were measured with microfluorometry.The peak of the emission spectrum
of DA fluorescence was at 490 nm.and of lipofuscin at 550 nm..The mean value
and SE of the intensity of adult rats was 55.813±3.302,and the aging was 28.905
±2.746(t test,P<0.001)respectively.This result suggested that the DA content
of aging rats was markedly decreased and its possible significance was discussed.
