人胎儿胸腺哺育细胞的组织化学和电镜观察 被引量:3

HISTOCHEMICAL AND ELECTRON MICROSCOPICAL OBSERVATION ON THYMIC NURSE CELL OF HUMAN FETUSES

摘要 本研究用组织化学法、扫描和透射电镜观察了人胎儿胸腺哺育细胞(thymic nurse cell,TNC)。结果表明,TNC 的 ANAE、AcP、AIP、ATPase、5′-Nase 和 PAS 均呈阴性反应,TNC胞质内含有数量不等的淋巴细胞,其中少数淋巴细胞的 ANAE、AcP、AIP 和 ATPase 呈阳性反应。TNC 内的淋巴细胞 ANAE 阳性率为26.8%。TNC 外的淋巴细胞 ANAE 阳性率为51.3%,两者的结果比较,有显著性差异(P<0.01,u=11.23)。扫描电镜观察,TNC 为球形,细胞表面较光滑,有少量短微绒毛。1μm 半薄切片显示,胸腺皮质内可见 TNC 样细胞,细胞界限清楚,核圆或椭圆形,染色淡。胞质中可见数量不等的淋巴细胞。透射电镜观察,TNC 胞质内可见张力丝,表明这些细胞具有上皮细胞特征。TNC 胞质内的淋巴细胞可见正常的线粒体、游离核糖体等细胞器。本文对上述结果与胸腺巨噬细胞作了比较和讨论。 This paper reports some histochemical,scanning and transmission electron mic- roscopical features of thymic nurse cell(TNC)of human fetuses.The results showed that TNCs were all negative for ANAE,AcP,AlP,ATPase,5'-Nase and PAS.TNC contained various numbers of lymphocytes.Some intracytoplasmic lymphocytes were positive for ANAE,AcP,AlP and ATPase.Their ANAE positive percentage deter- mined were 26.8%.However,ANAE positive percentage of extracytoplasmic lym- phocytes of TNC were 51.3%.Our data demonstrated that there was significant dif- ference between these two ANAE positive percentages,P<0.01(u=11.23).Under scanning electron microscope,TNC looked like somewhat smooth spheres with a few short microvilli.In 1 μm semithin section,TNC-like cells were located in cortical area of the thymus.The outline of the cells was clear,their nuclei were round or oval and stained relatively light.The cytoplasm contained lymphocytes.The transmission electron microscope observation showed tonofilaments in TNC and therefore proved its epithelial nature.Normal organelles such as mitochondria,free ribosomes etc were seen in the lymphocytes inside TNC.The above features of TNC were compared with those of macrophage.
