Effects of Adenosine Deaminase Activity on Human Sperm Motility

摘要 本研究发现人精子膜中有腺苷脱氨酶(ADA)活性,其Km值为266.94±1.31μM。比牛附睾尾部精子膜中ADA的Km高一个数量级。我们发现活力低于50%的不育者精子膜中的ADA活性(154.22±88.49 μM/mg/h,14例)明显高于正常人精子膜中的ADA活性(67.45±17.66μM/mg/h,25例),p<0.01。用ADA的特异性抑制物erythro-9-(2-hydroxy 1-3-nonyl)adenine(EHNA)处理洗过的正常人精子6 h后,实验组精子活力(91%)明显高于对照组(0%),p<0.01;同样条件下测得34例不育者精子的活力(83.86±11.21%)明显高于对照组(68.27±18.73%),p<0.01。另外EH NA处理后的精子,其穿透去透明带金黄地鼠卵的能力均有升高,正常人精子从54±8.66%升高到83±6.69%,不育者精子从21.33±55%升高到77±9.67%,p<0.01。 In this study adenosine deaminase (ADA) was found in human sperm plasma membranes. This enzyme has a relatively higher Km value of 266.94 ±1.31μM compared with that isolated from bovine caudal sperm. The activity of ADA in infertile human sperm (154.22±88.49μM/mg/h, n=l4) is significantly higher than that infertile human sperm(67.45±17.22 μM/mg/h, n=25) (p<0.01), A correlation was observed between the enzyme's activity and the sperm motility as well as the ability of sperm penetrating zonafree hamster egg. The motility percentage of 34 infertile samples (83.86±11.21%) treated with 14 μM is significantly higher than that of the control (68.27+18.7%). In addition, fertile and infertile human sperm's penetrating zona-free hamster egg rates were both significantly increased after the sperm was treated with 10μM EHNA (p<0.01). The data indicate that the activity of sperm-membrane-bound ADA affects human sperm physiological functions.
