摘要
通过诱导HU-TY-1(耐高温酿酒酵母)产孢及单孢分离得到的单倍体株HU-TY-1-A(Saccharomyces cerevisiae)与具有STA1、STA2、STA3(淀粉糖化酶)遗传因子的酵母(S.diastaticus)单倍体和纯合二倍体进行相同交配型的种间原生质体融合,融合率为3.3×10^(-6)—8.7×10^(-6)。融合子的细胞形态、大小、DNA含量、生长和发酵特性等方面均不同于亲株。融合株BA-2、BA-3、CA-18、DA-2、CCA-30、其细胞体积与DNA含量为亲株之和;30℃生长速率与生物量为亲株的2—3倍,40℃生长速率与生物量为亲株HU-TY-1-A的1.3—1.6倍,能够利用淀粉,并且在40℃高温葡萄糖发酵酒精产率比糖化酵母高,比HU-TY-1-A低,是一些有淀粉糖化酶、耐高温的酿酒酵母新菌株。
Novel S.cerevisiae BA-2,BA-3,CA-18,DA-2,CCA-30 were obtained by means of protoplast fusion, which processed as follows: The thermotolerant yeast(S.cerevisiae HU-TY-1-A)fused separately with S.diastaticus, such as the identical mating type auxotrophic haploids 5101-7C(arg-STA1) ,5206-1B(arg-STA2) ,and 5301-14D(leu-met-his-STA3) ,CC-1 (argSTA1/STA1) involving with 35% polyethylene glycol (PEG MW 4000) and 1mmol/L CaCl_2. The fusion freguency resulted from complementation of the parental nutrient markers is about 3.3—8.7×10^(-6). The comparative studies between the novel strains and the parental strains showed that their cell size, cell morphology, DNA content per cell, growth rate, biomass, fermentation ability and sporulating ability are quite different. The cell volume and DNA content of the novel strains are about the sum of that of two parents. The growth rate of novel strains at 30℃ and 40℃ is higher than that of parents. Their biomass at 30℃ is 2 times higher than that of parents. The biomass at 40℃ is 1.3 times higher than that of parent HU-TY-1-A. The novel strains are also capable of fermenting soluble starch and glucose at 33℃ and 40℃.
