摘要
本文探讨了海藻酸钠的结构组分对微囊强度的重要作用。提出了微囊化批量生产中估算活细胞的方法以及影响满囊比的主要因素等。制备微囊的成功率达到95%以上,满载率可达95%。微囊化杂交瘤细胞在静态培养6—10天后,细胞浓度为8.8×10~6~2.1×10~7cell/ml微囊。单抗浓度为379μg/ml微囊。微囊化产物经SDS电泳呈现一条或两条蛋白带,表明单抗纯度较高。
We reported here that constitute components of sodium alginate are very important factors affecting strenth of microencapsule. Method of estimating viable cells number in microencapsule in batch culture hybridoma has been developed and influence factors on the rate of full microencapsule. Now making microencapsules are very good. Rate of forming microencapsule and rate of full load was above 95%. Cell density of hybridoma were found to reach 8.8×10~6—2.1×10~7 cell ml^(-1) by the static culture for 6—9 days. McAb accumulated in the intracapsule to a final concentration of 319μg ml^(-1). Analysis of the intracapsule protein by SDS gel electrophoresis demonstrated that it was pure.
